Grape seed extract effective against cancer cells

A University of Colorado Cancer Center study published online ahead of print in the journal Nutrition and Cancer describes the laboratory synthesis of the most active component of grape seed extract, B2G2, and shows this synthesized compound induces the cell death known as apoptosis in prostate cancer cells while leaving healthy cells unharmed.

“We’ve shown similar anti-cancer activity in the past with grape seed extract (GSE), but now we know B2G2 is its most biologically active ingredient which can be synthesized in quantities that will allow us to study the detailed death mechanism in cancer cells,” says Alpna Tyagi, PhD, of the University of Colorado Skaggs School of Pharmacy and Pharmaceutical Sciences. Tyagi works in the lab of CU Cancer Center investigator and Skaggs School of Pharmacy faculty member, Chapla Agarwal, PhD.

The group has spent more than a decade demonstrating the anti-cancer activity of GSE in controlled, laboratory conditions. For example, previous studies have shown the GSE effectiveness against cancer cells and have also shown its mechanism of action. “But until recently, we didn’t know which constituent of GSE created this effect. This naturally occurring compound, GSE, is a complex mixture of polyphenols and also so far it has been unclear about the biologically active constituents of GSE against cancer cells,” Tyagi says.

Eventually the group pinpointed B2G2 as the most active compound, but, “it’s expensive and it takes a long time to isolate B2G2 from grape seed extract,” Tyagi says.

This expense related to the isolation of B2G2 has limited the group’s further exploration. So instead of purifying B2G2 from GSE, the group decided to synthesize it in the lab. The current study reports the success of this effort, including the ability to synthesize gram-quantity of B2G2 reasonably quickly and inexpensively.

In the paper’s second half, the group shows anti-cancer activity of synthesized B2G2 similar in mechanism and degree to overall GSE effectiveness.

“Our goal all along has been a clinical trial of the biologically active compounds from GSE against human cancer. But it’s difficult to earn FDA approval for a trial in which we don’t know the mechanisms and possible effects of all active components. Theore, isolating and synthesizing B2G2 is an important step because now we have the ability to conduct more experiments with the pure compound. Ongoing work in the lab further increases our understanding of B2G2′s mechanism of action that will help for the preclinical and clinical studies in the future,” Tyagi says.

Grape Seed Extract Bollixes Norovirus

In the study, the researchers observed that under treatment with grape seed extract, at low doses, the spherically-shaped murine (mouse) norovirus-1 coat proteins clumped, and showed "obvious deformation and inflation," according to the report. At higher doses, the researchers saw no coat proteins, only protein debris. "This provides evidence that [grape seed extract] could effectively damage the [norovirus] capsid protein, which could reduce viral binding ability and infectivity accordingly," according to the report.

The researchers used surrogate viruses because there are no suitable animal models of norovirus, and human norovirus has been impossible to propagate in cell cultures. Nonetheless, the researchers were able to measure the specific binding strength of human norovirus by two different methods, finding that it declined precipitously under the influence of grape seed extract, providing further support to their results.

Norovirus is transmitted mainly fecal-orally, and infected food handlers, contaminated water, and surfaces can be identified as important sources of transmission, which could further contaminate ready-to-eat foods, drinking water, shellfish, and fresh produce. A mere 10-100 virus particles are sufficient to transmit the disease.

An extract found in cinnamon bark, called CEppt has been shown to inhibit the development and progression of the disease in this latest study.

Researchers from the Department of Zoology at Tel Aviv University found that potent extracts from cinnamon bark inhibit the toxic amyloid polypeptide oligomers and fibrils that have been found in Alzheimer`s brain plaque formations. The healing power of cinnamon has been known since biblical times as high priests used the spice to protect against infectious disease. Antiviral properties have been confirmed by modern research, prompting studies to further examine extracts from the spice in the development of Alzheimer`s disease.

Scientists have isolated the CEppt active compound found in cinnamon bark and created an aqueous solution for use in research experiments. The solution was fed to genetically altered mice that have been predisposed to develop an aggressive form of Alzheimer`s disease. Many natural substances including resveratrol, curcumin and blueberries have shown promise in preventing this devastating form of dementia, but no therapy exists that can reverse the disease once a diagnosis has been presented. Professor Ovadia commented on the results of this research “The discovery is extremely exciting. While there are companies developing synthetic AD inhibiting substances, our extract would not be a drug with side effects, but a safe, natural substance that human beings have been consuming for millennia.”
 
Scientists conducting this study did not publish the amount of cinnamon used to produce their results. Nutrition experts recommend using cinnamon liberally sprinkled on food daily or supplementing with 500 mg of cinnamon bark extract taken daily with meals. Cinnamon is rapidly becoming a natural compound that may hold the key to prevent and treat Alzheimer`s disease.

Researchers publishing the results of two independent studies in the British Journal of Nutrition have demonstrated that polyphenol antioxidants found naturally in red grapes can prevent the buildup of fat in muscle tissue that is a precursor to metabolic disorder and diabetes. Resveratrol and grape seed extracts influence how the body responds to insulin, the hormone that controls critical mechanisms of dietary sugar and fat metabolism.

The grape-derived compounds have been shown to improve cardiovascular and Alzheimer`s disease risk factors in the past. Information gleaned from these studies explains how daily consumption of the fresh fruit can be invaluable as it prevents muscular fat accumulation leading to metabolic instability and diabetes.

High Sugar Diet Leads to Fat Accumulation in Muscle Tissue
Detailing the result of research performed at the University of Montpellier in France, scientists found that subjects fed a diet high in fat and sugar experienced a lower accumulation of fat in muscle tissue when supplemented with a concentrated extract of red grape polyphenols. Diets high in oxidized saturated fats are known to disrupt the normal function of insulin leading to insulin resistance and diabetes. The polyphenol may help to prevent cancer development, cardiovascular disease and dementia, and is now shown to impede the development of diabetes by regulating insulin sensitivity.

Resveratrol Improves Insulin Sensitivity to Prevent Metabolic Syndrome and Diabetes
Researchers formed two groups of participants and randomly supplemented their diet with resveratrol (5 mg twice a day) or a placebo. After four weeks the resveratrol group showed a significant decrease in insulin sensitivity compared to the placebo group. The study authors explain that the results are likely due to the potent antioxidant action of resveratrol, as oxidative stress is a key contributor to the onset of insulin resistance.

Researchers concluded “The present study shows for the first time that resveratrol improves insulin sensitivity in humans, which might be due to a resveratrol-induced decrease in oxidative stress that leads to more efficient insulin signaling.” To maximize the anti-diabetic health benefits of grape polyphenols, health-minded individuals will want to include organically grown red grapes in their diet or supplement with grape seed extract and resveratrol (10 to 25 mg daily).
